Botanical Description

Cinnamomum camphora, the camphor tree, is a large evergreen tree of the family Lauraceae reaching 20-30 m tall, with a broad, dense, dome-shaped crown and pale, vertically fissured bark. The alternate, glossy leaves are ovate to elliptic, 5-10 cm long, with three prominent veins arising near the base and a strongly camphoraceous scent when crushed; new growth flushes a bright pinkish-bronze. In spring it produces masses of tiny, fragrant, creamy-white flowers in axillary panicles, followed by small spherical drupes that ripen from green through red to glossy black. Native to East Asia (southern China, Taiwan, Japan, Korea, and the Ryukyus), it has been widely planted in subtropical regions worldwide for its timber and camphor and has become invasive in parts of Australia and the southeastern United States.

Camphor

Bicyclic monoterpene ketone

Concentration: Highly chemotype-dependent; ~37.8-84.1% of the essential oil in the camphor chemotype (up to ~93% in some leaf oils)

The characteristic constituent that gives the tree its name. Camphor produces a cooling-then-warming counter-irritant sensation on skin by activating TRPV3 and TRPM8 receptors, and has mild topical analgesic, antipruritic and rubefacient actions. Systemically it is a CNS stimulant and proconvulsant, which underlies the well-documented toxicity of ingested or over-applied camphor.

1,8-Cineole (eucalyptol)

Monoterpene oxide

Concentration: ~1-20% (dominant in the cineole chemotype; ~19.8% in wood oil)

An aromatic ether with expectorant, mucolytic and anti-inflammatory activity; it contributes the eucalyptus-like note of some camphor-laurel chemotypes and is the basis of their use in inhalant and chest-rub preparations.

Safrole

Phenylpropanoid (allylbenzene)

Concentration: Low in leaf/wood oil (0-3%) but up to ~57% in root-bark oil of the safrole chemotype

A hepatocarcinogen and hepatotoxin whose use as a food/flavour additive is banned in most jurisdictions. Its presence is the main safety concern of safrole-chemotype camphor oil and a reason root-derived oils are avoided medicinally.

Linalool

Acyclic monoterpene alcohol

Concentration: Dominant constituent of the linalool ('ho leaf/ho wood') chemotype (often >80%)

A floral-scented alcohol with sedative, anxiolytic and antimicrobial properties; the linalool chemotype of C. camphora is a major commercial source of natural linalool for perfumery and aromatherapy.

Borneol

Bicyclic monoterpene alcohol

Concentration: Minor to moderate; dominant in the borneol chemotype

A reduced relative of camphor with mild analgesic and anti-inflammatory action; borneol is valued as a penetration enhancer that increases transdermal and blood-brain-barrier passage of co-administered agents.

alpha-Terpineol

Monoterpene alcohol

Concentration: ~4-6% of wood and root oils

A lilac-scented alcohol contributing antimicrobial and mild antioxidant activity to the whole oil.

Nerolidol

Sesquiterpene alcohol

Concentration: Dominant in the nerolidol chemotype

A sesquiterpene alcohol with antimicrobial, antibiofilm and skin-penetration-enhancing properties, characteristic of one of the tree's less common chemical variants.

Limonene

Monocyclic monoterpene

Concentration: ~0.4-2.1%

A citrus-scented hydrocarbon contributing to the aroma and providing minor antioxidant and solvent-like properties.

⚠ Drug Interactions

Antiepileptic drugs / anticonvulsants

Major Evidence: Theoretical

Absorbed camphor is a CNS stimulant that lowers the seizure threshold and can precipitate generalized tonic-clonic seizures; this may pharmacodynamically oppose anticonvulsant control in susceptible patients.

Clinical note: People with epilepsy or on antiseizure medication should avoid ingesting camphor and avoid heavy or occluded topical use.

CNS depressants (sedatives, alcohol)

Minor Evidence: Theoretical

Camphor toxicity produces a biphasic CNS picture (initial stimulation/seizure followed by depression); concurrent CNS depressants could compound the depressant phase after a significant exposure.

Clinical note: Relevant only in overdose/accidental-ingestion scenarios rather than normal topical use.

Preparation Methods

Topical rub / liniment (external only)

Parts: Distilled essential oil (camphor), wood

Camphor is used externally in chest rubs, balms and liniments as a counter-irritant for muscle aches, minor joint pain, itching and to relieve cough congestion. In the United States the FDA limits over-the-counter camphor products to a maximum of 11%. Warning: never apply to broken skin, do not use under occlusion over large areas, and do not apply to infants or the faces of young children (risk of laryngospasm and systemic absorption).

Steam inhalation (cineole/camphor chemotype)

Parts: Essential oil

A few drops added to hot water for inhalation have traditionally been used to ease nasal and chest congestion. Keep away from the eyes; avoid in asthma where volatile oils may provoke bronchospasm; not for young children.

Aromatherapy / linalool-chemotype oils

Parts: Leaf (ho leaf oil)

Linalool-rich 'ho' oils are used in diluted aromatherapy and perfumery. Safety: camphor essential oil must never be taken internally; camphorated oil, camphor oil and camphor liniment are banned oral products, and ingestion is potentially fatal. Avoid all camphor products in pregnancy and lactation. Safrole-rich root oils should not be used medicinally because safrole is a hepatocarcinogen.

GC-MS characterization of camphor-tree essential oils identified camphor, 1,8-cineole, terpinen-4-ol, limonene and safrole as principal volatiles and demonstrated contact and fumigant insecticidal activity against stored-product beetles, supporting traditional use of camphor as an insect repellent.

Seizures due to high dose camphor ingestion

Reports in the poisoning literature document that ingestion of camphor-containing products causes rapid-onset seizures, with pediatric ingestions of 500-1000 mg associated with convulsions and death, informing the FDA restriction of over-the-counter products to <=11% camphor and the ban on camphorated oil.

Ayurvedic materia medica (karpura)

Classical and medieval India
Camphor (karpura) is used in Ayurveda as a cooling, aromatic drug for respiratory complaints, skin conditions and ritual purposes, and remains an ingredient of many topical balms.

European pharmacopoeias (Camphorated oil / Linimentum camphorae; British Pharmacopoeia)

18th-20th century
Camphor liniment and camphorated oil were official preparations used as rubefacients and 'stimulants'; their high poisoning rate later led to regulatory bans on the oral products.

Meduna's convulsive (camphor) therapy

1934
Ladislas Meduna used injected camphor to induce therapeutic seizures in psychiatric patients, an early convulsive therapy that preceded and inspired electroconvulsive therapy - a striking historical illustration of camphor's proconvulsant action.
